Description	The GFP-Expressing Rat Embryonic Stem Cells are established by electroporation of dark agouti rat stem cells with plasmid carrying CAG-EGFP. The cells are ubiquitous promoter-driven and green fluorescent protein (GFP)-tagged. The GFP- Expressing Rat ES cells could be useful in the field of developmental biology and in research studies assessing chimeric structures such as organs including the brain, liver, heart, kidney, thymus, and spleen in other organisms. This is done by injecting the GFP-Expressing Rat ES cells into blastocysts and tracking the development of the cells through fluorescence at any given time. The chimera forming ability was tested by injecting GFP-Expressing Rat ES cells into blastocysts of albino F344/N rat.
Quality Control	1) Alkaline phosphatase staining preformed with Histofine Fuchsin Substrate kit; 2) Puromycin selection with 1 µg/ml final concentration

Propagation	The base medium for this cell line is a mixture of 1:1 KnockOut^TM DMEM/F12 medium (Invitrogen; 12660-012) and Neurobasal medium (Invitrogen; 21103-049). To make the completed growth medium, add the following components to the base medium: 1 mM 2-mercaptoethanol (Invitrogen; 21103-049), 1% B27 (Invitrogen; 17504-044), 3 µM CHIR99021 (Axon; 1386), 1 µM PD0325901 (Axon; 1408), 10³ U/ml human LIF (Z101345), 1% L-glutamine (G275), and 1% Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
