Description	Luc-Akt-PH/YFP Stable MCF-7/B2 Stable Cell Line express the pleckstrin homology (PH) domain of Akt fused to Renilla luciferase with a YFP-Mem, allowing the ability to monitor the PIP₃ production in real time through the established BRET biosesensor pair. In which the Luc-Akt-PH will be recruited to the plasma membrane due to PIP₃ production which then results in an energy transfer between the luciferase and YFP. The cells are suitable for the screening and study of molecules on the PI3K/Akt pathway for diabetes and cancer-related research. This cell line is the can be used with the Luc-Akt-PH Stable MCF7 Cell Line (available at abm, Cat. T3160) for experimental studies.
Procedure Overview
Recommended Seeding Density	Thaw entire contents into an appropriate T25 flask as specified in the Propagation instructions.

Over Expression	Luc-Akt-PH through transfection of Luc-Akt-PH vector and selected with 1 mg/ml geneticin to derive stable cline which was then transfected with the pYFP-Mem vector and underwent selection with 1 mg/ml geneticin and 200 µg/ml hygromycin B
Propagation	Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow III medium available from abm (TM003). To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999) to final concentration of 10%, 1 mg/ml geneticin (G271), 200 µg/ml hygromycin B (G265), and Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
